Instrumentation

GEML operates specialized equipment for genome editing, sample preparation, quality control (QC), and sequencing. Some instruments are also made available for user-lab access through the external page b-fabric system.

external page 4D-Nucleofector

Delivery of molecules to manipulate biological systems has always been a challenge, with low efficiency and/or high toxicity being the main hurdles. Nucleofection nicely complements chemical and viral methods, by showing high efficiency in commonly used cell types and providing access to primary cells and difficult to transfect cell lines.

The Lonza 4-D nucleofector core unit controls nucleofection parameters and the handling of various functional units that support different nucleofection formats.

- external page X unit: large cuvettes and 16-well strips, for up to 100ul and 20ul volumes, respectively.

- external page Y unit: 24-well culture dish format. Compatible with nucleofecting cells in adherence.

- external page LV unit: Large Volume setup for the nucleofection of up to 1xE9 cells.

Optimizing nucleofection conditions for your favorite cell type is not a trivial, but should also not been seen as an impossible, task. Please note that we do not offer user access to the 4D-nucleofector. Such experiments, typically part of larger collaborative projects, are conducted by our team.

96 well add on

external page 96-well Shuttle

This 4-D nucleofector add-on is ideal for higher-throughput experiments using the 96 well format, for example when transfering arrayed CRISPR libraries, during secondary screens.

Furthermore, implementing matrices of conditions facilitates assay establishment for difficult to transfect cells.

The focused-ultrasonicator allows the sonication and fragmentation of samples in an accurate and reprodicible manner. The LE220 supports 96well plates, allowing parallel processing of samples.

Two thermal cyclers with 96-deep-well blocks to support NGS library preparation and ddPCR applications.

Microfluidics platform that can generate barcoded partitions for downstream single-cell analysis applications.

The digital droplet PCR (ddPCR) quantifies nucleic acids using clonal amplification within discrete partitions inside each reaction.

The fragment analyser utilizes capillary electrophoresis for the parallel separation and analysis of up to 48 samples.

This alternative to a UV spectrometer, the Qubit fluorometer can accurately measure DNA, RNA, and protein concentration with high sensitivity.
